Application of Fluorescent Method to the Analysis of Vitality on Single Cell Models of Diatom, Horsetail and Fern

The chlorophyll autofluorescence observed with luminescence microscopy technique, including laser-scanning confocal microscope and microspectrofluorimeter, have been applied to the vitality determination of model cells such as diatom algae Ulnaria ulna, horsetail Equisetum arvense and fern Asplenium scolopendrium. In every cell population, it was possible to differ living, dead and stressory individuals. Their fluorescence spectra recorded by microspectrofluorimeter showed the chlorophyll maximum at 680 nm in living cells and peak only at 520 nm in dead ones. The method may be useful for laboratory practice because permitted to differ living and dead or undeveloped cells in the population as well the level of their vitality under a storage.
